This one suggest (page 12) that 3, 4 is connected to OpenTherm 3.0., meanwhile 5, 6 is connected to the outside temperature sensor on that block. That numbering next to the block is not matching the X3 numbering. You need to follow the instructions and check how the wires are running to the X3 terminal.

About removing the jumper, this is what I found in a different manual:

There are two versions of that manual, @kamilstan read the newer, which have an 8 pin connector for connecting external devices, meanwhile on his picture, there is a 6 pin connector for this purpose, included in the old manual which I linked. Brotje might have done a bit of upgrade on that.
Surprisingly these quite new (meaning few years old) Brotje heaters have quite old setups, not supporting the BSB standard, which would give more control over the heater.
